The framework in India
India has no biobank-specific law; biobanking is guided by ICMR's National Ethical Guidelines for Biomedical and Health Research Involving Human Participants (2017), which cover consent, governance and the need for detailed SOPs for collection, coding, storage and sharing. ICMR has also issued guidelines for establishing biorepositories within its own institutes.
Internationally, ISBER — the International Society for Biological and Environmental Repositories — publishes Best Practices: Recommendations for Repositories, now in its fifth edition (2023), which many biobanks use as their operational reference, including on backup storage and environmental monitoring.
Match the temperature to the material
The right tier comes from the material, the intended analyses and the protocol — not from a habit of putting everything in the coldest freezer. Over-specifying wastes energy and capacity; under-specifying can ruin a study.
| Tier | Typical equipment | Often used for |
|---|---|---|
| +2 to +8 °C | Laboratory refrigerator | Short-term holding, some reagents and fixed tissues per protocol |
| −20 °C class | Laboratory freezer | Some purified nucleic acids and reagents |
| −60 to −80 °C class | Ultra-low temperature freezer | Serum, plasma, many tissues and extracts for long-term storage |
| Below −150 °C | Liquid nitrogen vapour phase | Viable cells and materials that need cryogenic storage |
Inventory is a temperature control
Every minute spent searching for a box with the freezer door open warms everything near the door. A sample inventory — freezer, shelf, rack, box and position for every tube — is therefore part of temperature control as well as data management. Use labels that survive deep cold and frost, and keep a printed rack map on or near each freezer for when the system is down.
Continuous monitoring
- An independent probe in each freezer, separate from the freezer's own controller, with probes rated and calibrated for the temperature being measured.
- Alarms that reach a person round the clock by SMS, call or app, with an escalation chain if the first person doesn't respond.
- Battery backup for alarms and loggers so a power cut doesn't silence them.
- Daily review of trends; a freezer that takes longer to recover after openings is often warning of a failing seal, dirty condenser or ageing compressor.
- Records kept for the life of the samples, not just a few months.
Backup capacity
When an ultra-low freezer fails, its contents need somewhere to go within hours. Repositories keep spare capacity in an empty or partly empty freezer at the same temperature, sized to take the contents of the largest unit. Without it, a single compressor failure becomes a crisis.
Practise the move: who is called, where the spare keys and gloves are, how boxes are carried, and how the inventory is updated afterwards.
Site risks in Indian institutions
- Heat: rooms full of ULT freezers become very hot without ventilation or air-conditioning, shortening compressor life. Keep within the rated ambient.
- Dust: clean condenser filters often, especially in older buildings or near construction.
- Power: give each freezer its own earthed circuit, voltage protection and generator or UPS cover sized for compressor start-up.
- Monsoon humidity: more frost on doors and gaskets; increase gasket checks during the wet months.
- Weekends and holidays: make sure alarm escalation works when the building is empty.
Maintenance and replacement
Plan preventive maintenance — filters, gaskets, alarms, controller checks — and track each freezer's age and service history. Ultra-low freezers have a working life, and replacing one on a schedule, with samples moved calmly, is far better than replacing it after a failure at 3 a.m.
Frequently asked questions
- Should all biobank samples be stored at −80 °C?
- No. The right temperature depends on the material and its intended use. Follow the protocol for each collection; some materials need cryogenic storage, others are fine at higher temperatures.
- How much backup freezer space does a biobank need?
- Enough to take the contents of at least your largest freezer at short notice is a common planning principle. Check ISBER's best practices and your institution's policy for your situation.
